Abstract
Aim of this bachelor thesis was to find the impact of different preparation of moss samples on the ZnO nanoparticles accumulation and cell membranes disruption. Pleurozium schreberi Brid. Mitt. moss samples were gathered, treated in various ways under laboratory conditions and exposed to ZnO nanoparticles. Sequential elution technique was used to determine the concentration of Zn and K in four parts of the thallus – on the surface, in extracellular and intracellular spaces and in the residual material. Cell membrane disruption was evaluated by K concentration ratio inside/outside the cell.
It was discovered that washing the moss in tap water, regular wetting and placing the samples into dryer at temperature 50 °C negatively affects cell membranes integrity. Thus, it is not recommended to use these treatments to prepare samples used in nanoparticles biomonitoring.
