Vývoj LC-MS/MS metody pro látky ze skupiny sartanů a jejich odbourávání vlivem fotokatalýzy

Abstract

The diploma thesis is focused on the development of an analytical method of high-performance liquid chromatography with tandem mass spectrometry (LC-MS/MS) for the determination of selected antihypertensives from the group of sartans. This method was subsequently used to monitor the decrease in the concentration of the antihypertensive eprosartan mesylate during photochemical and photocatalytic degradation. As part of the method development, a suitable analytical column and mobile phase for liquid chromatography were selected and the mass spectrometer parameters were optimised. The best results were achieved on a Kinetex Phenyl-Hexyl column, with a mobile phase composed of an aqueous solution of 0.1% formic acid with 10 mM ammonium acetate and acetonitrile. For the mass spectrometer, an electrospray with an ionization voltage of 5 500 V and a drying and nebulizer gas temperature of 350 °C was chosen as the ionization source. For the photodegradation experiments, an eprosartan mesylate solution with a concentration of 10 μg cm-3 was chosen. During the first 30 min of the experiment, a decrease in analyte concentration to almost zero values could already be observed.
