Genotypové a fenotypové vlastnosti bakteriálního kmene Rhodococcus degradans a možnosti jeho biotechnologických aplikací v metodách biotěžby kovů

Abstract

It is known, that bacterial communities constitute an important part of the biosphere of the Earth, but their consistency and generic variations are little explored. Dissertation engages in genotypic and phenotypic characteristics of the bacterial strain Rhodococcus degradans. The practical part is focused on the basic characteristics of the bacterial strain Rhodococcus and then on reactions of the bacteria to the environment with metals (biosorption, bioaccumulation, biovolatization). The experimental part is divided into four main chapters (morphological properties of Rhodococcus degradans, genotypic, phenotypic and other physiological characteristics of the strain), results and discussion. Basic genotyping of the strain was carried out with use of the universal primers of the 16S rRNA. Phenotypic properties were detected with use of modern testing BIOLOG MicroStation system and were compared with the closest types within so far known taxonomic position of the strain. Resistance, biodegradation and bioleaching studies are ranked among other physiological characteristics. In conclusion there is given a summary and outline of the possible use of the bacterial strain.
