Abstract
The dissertation deals with the present situation of the territorial background of the environmental burden after mining of antimony ores – sludge lagoon Poproč (Slovak Republic) with using microorganisms as indicators of the current state of the environment. The aim of the dissertation thesis was to evaluate the current level of contamination of the affected area of the sludge lagoon and the degree of environmental toxicity using the characteristics of microbiota and necessary analyzes of its condition. The theoretical part of this thesis focuses on the role of autochthonous microflora in the environment after the extraction of polymetallic ores; deals with the useful role of microorganisms on reducing concentrations of toxic metals and metalloids in the environment and its important part in important part in renewal the original function of the affected environment. Experimental part of the thesis contains of physical-chemical’s characteristics analyzes in the substrates of the sludge lagoon. Biological characterization was determined on the basis of methods for determination of soil microbial biomass, microbial activity and metabolism including verification of abundance and diversity of indigenous microorganisms. To verify the biotransformation activity of the microbiota in the release of antimony and arsenic in the technogenic substrates of the sludge lagoon, heterotrophic bioleaching experiments were used in which the obtained isolates of bacteria genus Bacillus and microscopic filamentous fungi Aspergillus were applied. A state of the enviromental burden was defined on the basis of the results to predict further progress and the gradual revitalization of the area of interest was characterized.
