Abstract

This diploma thesis focuses on methods High Performance Liquid Chromatography (HPLC) and Liquid Chromatography with Mass Spectrometry (LC-MS/MS) which are used to determine busulfan during the treatment of patients with acute myeloid leukemia (AML). The aim of this work is to compare these methods HPLC and LC-MS/MS for clinical use. First part of the thesis describes diagnostics of AML and issues related to its treatment, namely metabolism of busulfan, its pharmacodynamic and pharmacokinetic relationships, methods, and the principles of HPLC and LC-MS/MS methods. Practical part focuses on determination of busulfan by HPLC and LC-MS/MS, which is necessary for the treatment of patient with AML. This part describes the preparation of reagens, samples, and also the chromatographic conditions for both methods. After the preparation phase the levels of busulfan were determined in 204 patients’ samples by both methods HPLC and LC-MS/MS. The statistical evaluation was done using MS Excel and Statistics 7. In conclusion, advantages and disadvantages of both methods in term of analytical performance and clinical benefits were evaluated.
