Abstract
Aquaponic system is an interconnection between aquaculture and hydroponics. In aquaculture, fish produce nitrogenous waste material and water from the tanks must be cleaned. In hydroponics, plants must be supplied with expensive nutrient solutions. The aquaponics combines fish farming with plant cultivation to solve the above problems. An important step is to check the quality of water and the content of dissolved nitrogen substances. The newly used method could be surface plasmon resonance (SPR), which uses a flow measurement system. The aim of the thesis was therefore to design and test biochips suitable for detection of nitrogenous substances in water. Ultraviolet and visible spectrophotometry was used as the comparison method. Two biochips have been prepared to detect nitrogen compounds in model waters. In the experimental part, the influence of the matrix, which is not negligible in real samples, was evaluated. As the solution, to use a two-channel SPR was proposed, whereby a binding is monitored on the first chip, and the matrix of the sample is subtracted in real time from the second chip.
