Abstract
This diploma thesis deals with biochemical measurements using Surface Plasmon Resonance imaging (SPRi). The aim of the work was to find a suitable reference substance serving as a negative control. Finding this substance would allow detecting lower concentrations of the analyzed proteins in the solution. The reference substance was searched between a selected antigens. Serum albumins from various animal species have served as analytes for the purpose of the experiment. The individual concentrations of analyzed albumin in solution were 100, 10 and 1 mg / l. Two SPR biochips were designed and prepared in the work. Biochemical measurements were performed on these biochips. At the same time, AFM atomic force microscopy was used to study the golden surface of the SPRi biochip.
